Hi. According to BHphoto YES. I do not possess an F5.

Q: will this work on nikon d3100

It will not work on d3100. It only works on D700,D800 or D4 which have a round view finder

Q: I often shoot with manual lenses, has anyone found that this helps with focusing manually? I often disregard the focusing dot with manual lenses.

Provided your diopter is set correctly, it helps. You can still see the entire focus screen, I’m not sure what AstroBob is talking about. However, manually focusing is still a skill in its own, and buying an accessory won’t make you a master overnight.
