Work on a tele with a Bigsby B5? Looking to eliminate the grinding sound of the strings on brass saddles.

They probably would but I would sooner do some file work on your existing brass saddles first.

Do these need to be slotted before use?

on tele saddles i do not slot. i do make sure the string spacing is correct before i bring the strings up to full tension. i have found with graph tech and brass saddles the strings cut their own slots.

Will it fit an American special telecaster?

Yes. Any of the 3 saddle bridges. You'll definitely get better intonation. Have these on all of 3 saddle teles.
