Q: Will this work with the canon m50 mark ii with a sigma 16mm lens (or canon ef-m 22mm lens) without causing a vignette view?

My camera is Canon M50 too. There are different sizes of adapters along with the package to fit your lenses.like 45 mm, 49mm, 52mm, 55mm, 62mm, 67mm,72mm and 82mm. I guess you can order separately for sizes that are not included in the package.

Q: Can the dsrl camera be placed vertically and the edges of the teleprompter do not affect vertical vision?

Yes, you can place the camera vertically. This teleprompter will be hung on your dslr camera lens through the lens adapter ring. You just need to check whether your camera lens size is match with one of the included adapter ring 49/52/55/58/62/67/72/77/82mm.

Q: Can it have a rode mic mounted in the dslr?

Yes but you would have to purchase an additional mount to make it happen. I usually just mount my Rode mic on a second tripod directly in front of the teleprompter.
